Pro-and Antioxidants in the Field of Fats XVIII: The Effect of Iron-porphyrin-compounds on the Autoxidation of Polyenes 111
The influence of anti-and prooxidants on the inversion of the catalytic effect of hemoglobin and cytodrorne c in the autoxidation of linoleic acid in aqueous phosphate buffer medium was investigated. B y addition of an antioxidant (thymol, a-tocopherol) or a second invertible prooxidant (Fe-0-phenanthroiine) the ,,critical catalyst concentration" at the inversion point can be lowered. Die ,kritische Katalysator-Konzentration" der Hamin-Verbindungen, d. h. die Konzentration des Katalysators, bei der Inversion von pro-zu antioxydativer Wirkung eintritt, w i r d , wie f r u h e r ausgefiihrt ', durch Herabsetzung des Peroxydgehalts des Substrats erniedrigt. Der gleiche Effekt kann durch Einsatz einer relativ kleinen Menge eines geeigneten A n t i o x y d a n s oder auch durch Zusatz eines zweiten P r o o x y d a n s erreicht werden. Hieriiber sol1 im folgenden berichtet werden. T e s t s u b s t a n z e n Hamoglobin (Hb) wurde wie fur die fruheren Versuche' aus Schweine-Erythrocyten hergestellt, Gehaltsbestimmung s. dort. Cytochrom c aus Pferdeherz entsprach ebenfalls dem fruher eingesetzten Praparat. Eisen(I1)-o-phenanthrolin: Aquivalente Mengen (1 : 3) von Eisen(I1)-sulfat p. A. (E. Merck, Darmstadt) und o-Phenanthrolin p. A. (E. Merck, Darmstadt) wurden zusammen in Wasser gelost. Thymol wurde als 0.05°/oige wadrige Losung von Thymol krist. DAB. 6 (E. Merck, Darmstadt) angewendet. D,L-a-Tocopherol N. F. XI (E. Merck, Darmstadt) wurde in absolutem Athanol p. A. (E. Merck, Darmstadt) gelost und rnit absolutem Athanol jeweils soweit verdunnt, dad die erforderliche Menge als 0.05 ml alkoholische Losung dem Versuchsansatz zugesetzt werden konnte. Die Gehaltsbestimmung erfolgte durch Extinktionsmessung bei 292 mp. Ascorbinsaure wurde als n/10 wadrige Losung von Natrium-L( +)-axorbat (E. Merck, Darmstadt) eingesetzt.
