✦ Synopsis


A chemical degradation and radioassay procedure is described for jinding the radioactivity in each carbon-atom of carbon-I4 labelled propionic and n-butyric acids. This method has been tested with suitably labelled compounds and found to be isotopically reliable.

INTRODUCTION.

The Schmidt (1-8), and Roseman (lo) reactions have been most frequently used for the complete degradation of carbon-14 labelled carboxylic acids (I1, 12). However, each of these methods has some limitations concerning the compound to be degraded and the yields and radiochemical purity of the intermediate degradation products.

A simple and rapid procedure for the determination of the millimolar radioactivity associated with each carbon atom in carbon-14 labelled propionic and n-butyric acids is presented. This has been accomplished by using the modified Hunsdiecker reaction in combination with the C-methyl oxidation procedure.

DISCUSSION.

This paper describes the development of an isotopically reliable procedure for the complete chemical degradation of carbon-14 labelled propionic and n-butyric acids on a small-scale. In addition to the carbon-14 labelled acids degraded in the previous paper (I3), propionic-3-14C and n-butyric acid-2-14C have been employed as model compounds to test this process.
