Model-free sib-pair linkage analysis was used to screen 367 highly polymorphic markers for evidence of linkage to a disease, defined either quantitatively (Q1) or dichotomously (AF). Five individual replicates, plus a case family data set containing all families in these replicates with at least one

β¦ Synopsis
Sib-pair linkage analyses were used to search for linkage to a set of chromosome 19 and 21 marker loci in two sets of families with Alzheimer's disease. The advantage of this technique is that no assumption is made about the mode of inheritance of the disease. Some mild suggestions of linkage were found in earlyonset families for a chromosome 21 marker and in a set of late-onset families for a chromosome 19 marker. CD 1993 Wiley-Liss, Inc.

## Abstract An improved sibβpair test for linkage is introduced which is superior to the previously proposed tests. The test is derived from the standard chiβsquared goodness of fit statistic by restricting the alternative hypothesis to the genetically possible. Critical values are given and exact
Nick examines the problem of testing for linkage between a disease susceptibility locus and a marker locus for sib-pair data. Given a specified simple alternative for the parameters (pO, p I , p 2 ) of the multinomial distribution of (NO, N , , N2), where Nj denotes the number of sibs sharing exactl
Four relative-pair methods for detecting genetic linkage were applied to familial Alzheimer's disease data. Results obtained using an extended Haseman-Elston test and a weighted rank pairwise correlation test, which both use information from all relative pairs, were consistent with previously publis
Sib pairs drawn from the simulated common oligogenic disease families were selected for extreme quantitative trait scores and analyzed using interval mapping and multipoint methods. Linkage analyses of 112 selected sib pairs, in which one or more members had trait values exceeding the disease thresh
