✦ Synopsis

Propylproline and N-demethyl-lincomycin, two i n t e r m e d i a t e s i n l i n c o m y c i n b i o s y n t h e s i s , were prepared i n r a d i o l a b e l e d form.

[**C]-labeled N-demethyllincomycin was s y n t h e s i z e d by a r e s t i n g cell suspension of a lincomycin n e g a t i v e mutant o f Streptomyces l i n c o l n e n s i s t h a t was f e d methylthiolincosaminide and uniformly labeled [ '*C]-tyrosine.

[ '*C]-N-demethyl-lincomycin was isolated from t h e C u l t u r e medium by ion exchange chromatography and r e v e r s e phase-ion p a i r h i g h p r e s s u r e l i q u i d chromatography.

[**C]-propylproline was generated by t h e base h y d r o l y s i s of [ "C]-N-demethyl-lincomycin and isolated by anion exchange chromatography. The products had radiochemical p u r i t i e s g r e a t e r than 95% and s p e c i f i c a c t i v i t i e s o f 5.9 mCi/mmle.

## Abstract ^14^C‐Labelled vincristine was prepared from N‐desmethyl vinblastine by formylation with ^14^C‐formic acid. Excess radioactive formic acid was recovered as sodium formate. ^14^C‐Labelled N‐desmethyl‐N‐formyl‐leurosine was prepared analogously.
