β¦ Synopsis
We present a simple method for determining the stoichiometry of three different proteins which had been radioiodinated using two different labeling agents. The method consists of developing a silica gel thin-layer chromatography plate containing an aliquot of the crude reaction mixture in ethanol and subsequently measuring the radioactivity at the origin. This procedure obviates the need for prior purification of the protein, requires less protein than most other methods, yields a precise stoichiometry, and is particularly useful for time-course studies. The method may be applied to the quantitative analysis of other macromolecules which have been derivatized by alcohol-soluble reagents.
