✦ Synopsis

A novel route for the chloromethylation of polystyrene grafted Multipin supports (Synphase ru Crowns) has been developed Aminomethylated polystyrene was converted into chloromethylated polystyrene via a simple diazotisation reaction. The modified graft polymer was assessed by solid phase synthesis of a model dipeptide.

A new aminoethyl-polystyrene linker, stable at low concentrations of TFA, has been developed for the solid phase synthesis of peptide amides. The described linker is stable under conditions which remove Bu(t) protecting groups (30-50% TFA in DCM) and the desired product can be finally cleaved off th

A simple linker has been developed for the attachment of ahtehyde fimctionality. The aldehyde substrate is condensed with a support-bound serine or threonine to give an oxazolidine. The oxazolidine linker is stable to the conditions of Fntoc peptide synthesis, inchtding TFA treatment, but is cleaved

A new and simple linker has been developed for synthesis of secondary amides and sulfonamides. This application is based on the reductive amination reaction between the acetophenone subunit of the linker and a wide range of primary amines including aliphatic and aromatic amines. We have developed a useful method for the loading of various nucleophiles such as anilines and phenols onto Multipin Crowns containing the Rink linker, which cannot be readily attached by other methods. The product of the reaction was confirmed using Magic Angle Spinning NMR.
