Abstract
Radiolabelled neurokinin‐1 (NK~1~) receptor antagonists 1b and 2b were required for in vitro/in vivo characterization to support the development of 1a and 2a as fluorine‐18 labelled PET ligands. These tritium labelled compounds were synthesized from aryl iodide precursors giving the final tritiated tracers with specific activities of 28 (1b) and 14 (2b) Ci/mmol. The synt esis of 3H-labelled azidothymidine (AZT) from accomplished by column chromatography. The purity of the labelled compound was determined by high performance 1 iquid chromatography connected with a radioactive flow detector. The labelled AZT was found to be stable at pH ranging from 2.3 to 7.
S)-3-Aminoquinuclidine-3H 1 Oc-S having a specific activity of 66 Ci/mrnol was prepared in eight steps from lsonicotinic acid (2). Reduction of 2 with carrier free tritium gas over PtO2 In DMF gave isonipecotic acid-3H &. Conversion to a-bromo ketone followed by cyclization afforded 3-quinuclidone-
R) and (S)-3-aminoquinuclidines-3H with the specific activity of 35 Ci/mmol were prepared. Reduction of enamide 2 with carrier free tritium gas over RhCODCl dimer, (2S, 3s) Chiraphos in methanol gave yacemk amide &. Hydrolysis followed by resolution of the enantiomers with (R)-methyl benzyl isocyana
