The eyntheeie of 2-chloro-10-(3'-dimethylaminopropyl)phenothiazine-3', 3'-d2 (Chlorpromazine-d2) and ite four major metabolitee (the mono-and didemethylated aminee, chlorpromazine sulfoxide, and chlorpromazine N-oxide), all containing the 3',3'-d2 label in > 95% ieotopic purity, ie deecribed. Considerations leading to the optimum rider and poeition of the labels are diecuaeed, and the problem of coupling the aliphatic aide-chain to the tricyclic eyetem ie avoided by utilizing the readily available Z-chloro-10-(2'-cyanoethyl)phenothiazine as starting material, 80 that the carbon skeleton of the aide-chain ie already in poeition before the ieotope ie introduced.
