The first total synthesii of the highly unstable biological medistor 12-ket&coss&etr~ic acid (12-KBTB) 3 and its 89-traas -isoma tp is presented. The strategy focuses on the stable precursor dithiane u and its ccnwzsion to gand 211. Bicchemical experiments show that the two isomers are net interccmveited in vivo. raising the possibility that the rrans-isomer Zp may be formed by a pimary biihemicsl mechanism.
