We report the synthesis of 1,4-dihydropyridines on solid support utilizing the Hantzsch condensation. Unwanted reactive impurities were removed from the desired products, by selective cleavage from solid support. The synthesis of a 272 compound library is reported using the methods described.
Tritium-or 14C-labelled methyl iodide and ethyl iodide were used for alkylations of phenols and thiols. The reactions were performed in a two-phase system with phase transfer catalysis at room temperature. Products of high specific activity were isolated in isotope yields ranging from 38% to 100%.
