β¦ Synopsis
The present phosphotriester strategy for the (solution-phase) synthesis of the 3'-5' internucleotide bond of DNA is basically a two-step process' (Fig. ). We have modified and simplified this process by coupling together these up-to-now generally uncoupled reactions* into one continuous and abbreviated reaction sequence (Fig. ). These modifications lead to an increase in the overall yield to near quantitative as well as to a decrease in the working time by at least a factor of 4.

Most of the published work on the chemical synthesis of oligonucleotides\* has involved the us8 of intermediates with unprotected lnternucleotide phosphodiester linkages. Such intermediates can usually be isolated only on a comparatively small scale and are sometimes difficult to obtain pure. Furthe
## Abstract The synthesis of a total of twelve DNA fragments on the solid supports cellulose or polystyrene via a phosphotriester approach is described. Dimer and trimer blocks, protected at the 5β²βend with the levulinoyl group, were used for chain extension in the presence of the activating agent
A novel method for the efficientsynthesis phosphorodithioate DNA using H-phosphonothioate synthons is described. The 5'-DMT-nucleoside H-phosphonothioate monomers were prepared in two high yielding steps from commercially available phosphoramidites. This strategy for the synthesis of Hphosphonothioa
