✦ Synopsis

Reduction in alkaline phosphatase activity was observed when HeLa S3 cells were grown in Puck's medium containing high concentrations of human serum. This effect was not seen with the enzyme of Chang liver 8A cells. The induction of increased alkaline phosphatase in HeLa S3 by prednisolone or by osmolality changes was not prevented by serum. The concentration of serum in the culture medium had no influence on acid phosphatase activity.

## Abstract Parallel changes in the enzyme activities of Ca^2+^ ATPase and alkaline phosphatase were observed in HeLa cells. Both enzymes were inhibited to a similar degree by L‐phenylalanine, L‐tryptophan, and L‐leucine, while being relatively resistant to L‐homoarginine. Exposure to heat (56Β°C, 6

HeLa (substrain Ho) grown in serum free medium showed an increase in the specific activity of alkaline phosphatase when fetal calf serum (10%) was added to the medium (9.7 nmoles/sec/mg protein to 86.8). There is ample evidence of genetic differences in the level of alkaline phosphatase in blood serum or plasma. Differences have been described between strains of rats (Tuba, '53), and breeds of cattle Kunkel et al., '53; Rusoff et al., '54) and swine (Combs et al., '59). Hypophosphatasia in humans ha

The techniques for measurement of biosynthetic rates and intracellular transit times of an integral membrane protein isoenzyme have now been validated. Thus, induction of placental alkaline phosphatase in cultured HeLa cells by prednisolone and by butyrate is shown to result in its increased biosynt
