✦ Synopsis


A rapid micro method of preparing FAME for GC analysis of oils containing CLA A rapid micro method of preparing FAME for GC examination suitable for oils containing conjugated linoleic acids (CLA) is described. This involves base-catalysed methanolysis but avoids the late acidification step in similar procedures which would isomerise esters with conjugated isomerisation. A mixture of toluene and methanolic sodium methoxide was added to samples containing CLA mixed with an internal standard (methyl heptadecanoate). This mixture was heated to 508C in a water bath for 10-12 min and then cooled for 5 min. After addition of acetic acid and water, esters were extracted with hexane and dried with sodium sulfate. This procedure requires 30 min.
