The amount of free e-amino groups of lysine residues in proteins can be determined by a fast and straightforward method based on the use of r9F NMR spectroscopy. Unsubstituted e-amino groups are trifluoroacetylated with the reagent, S-ethyl trifluorothioacetate, in dimethyl sulfoxide solution, and t
