✦ Synopsis
The amount of free e-amino groups of lysine residues in proteins can be determined by a fast and straightforward method based on the use of r9F NMR spectroscopy. Unsubstituted e-amino groups are trifluoroacetylated with the reagent, S-ethyl trifluorothioacetate, in dimethyl sulfoxide solution, and the number of such groups is quantitatively determined using standard 19F NMR techniques. The method is evaluated using the following proteins: a-lactalbumin, P-lactoglobulin, bovine serum albumin, p-casein, and lysozyme.
