โœฆ Synopsis


Autophagy, a tightly regulated homeostatic pro-survival mechanism, has also been implicated in pathogen recognition and clearance by macrophages as part of the innate immune response. In the May issue of EJI, Mitroulis et al. shed light into the role of autophagy in human neutrophil biology, associating this process with oxidative burst and pathogen phagocytosis. The authors demonstrate recruitment of the neutrophil autophagic machinery after rapamycin-induced mTOR inhibition or selective activation of Toll-like receptors (TLR). Phagocytosis of Escherichia coli by neutrophils leads to a similar induction of autophagy in a phosphatidylinositol-3 kinase-and reactive oxygen species (ROS)-dependent manner. Interestingly, IL-1b, but not TNF-a, activate the autophagic machinery. This novel report provides a springboard to further delineate the role of autophagy in neutrophil-driven inflammatory responses and assess the potential interplay between autophagy, cell survival, and inflammation.
