✦ Synopsis

To investigate in-vivo concentrations of cytoplasmic phosphate, especially during dynamic conditions, a method has been developed that enables reproducible determination of cytopiasmic phosphate from 5 pM up to 30 FM. The method involves fast sampling. spontaneous inactivation of cell metabolism and differential extraction procedure to gain porosity of the outer cell membrane exclusively. To determine very tow cytoplasmic phosphate concentrations, an enzymatic assay was linked to a sensitive spectrophotometric cycling method to increase the detection limit.

We describe a modified colorimetric method that quantitates inorganic phosphate linearly up to 60 nmol, with high stability of the developed color and with a low interference by ATP concentration (up to 30 mM). This method is very suitable for use in ATPase enzymatic assays, especially with enzymes

Substrate oxidation by various organs in animals as well as in humans is usually studied by experiments in which radioactively labeled substrates are used and the production of is measured. In vivo, substrate oxidation by 14CO 2 an organ has, up to now, not been determined by means of stable isotope
