✦ Synopsis

An efficient synthesis of protected ribonucleosides useful for phosphoramidite RNA synthesis is described. Di-t-butylsilylene group was employed for simultaneous protection of 3%-and 5%-hydroxyl functions of nucleoside. Subsequent silylation of free 2%-OH group followed by introduction of suitable protection on the base moiety, removal of cyclic silyl protection and tritylation of 5%-OH gave target compounds in 60-66% overall yield.

We report a new method for protecting nucleosides for hydrogen phosphonate RNA synthesis that gives very high yields for the purines. This method is based on our finding that, for adenosine and guanosine, silylation of a mixture of 2' and 3' H-phosphonates in the presence of DBU is highly selective

The synthesis of atlyl-and allyloxycarbonyl (AOC)-protected RNA phosphoramidites is reporlexl. The use of allyl and AOC groups allows the chemistry of solid-phase RNA synthesis to be expanded to include base-labile modified nucleosides, such as acetylcytidine. The allyl-and AOCprotective chemistry c
