✦ Synopsis


A sensitive assay for 5'-nucleotidase activity is described. The method involves incubation with radiolabeled [3H]adenosine 5'-monophosphate (AMP) and thinlayer chromatography on polyethyleneimine-coated plastic to achieve separation of substrate (AMP) from product (adenosine). The resulting spots can be excised and counted in a scintillation counter to give a quantitative determination of 5'-nucleotidase activity. The method allows detection of less than 1.5 x 10e5 units of enzyme.
