✦ Synopsis

A new reagent --4-nitro-l-H-pyrazole-l-[N, N'-bis(ten-butoxycarbonyl)]cmboxamidine --has been develolxd to effect the rapid a~l efficient synthesis of I~(~)-pmtected guanidin~ from and secendm3, amines. The reagent is a more eleclrophilic, sad comfy more derivative of the literature reagent l-H-pyrazole-l- [N, N;bis(tert-butoxycmbonyl)]catboxmrdcfine. The increased reactivity of the new reagent affords it inacased yields in solution mid the need for fewer equivalents when guanylating resin-bound amines.

The clean and efficient cleavage of N-benzyl linked tertiary amines from a solid support (e.g. 4) by treatment with ct-chloroethyl chloroformate (ACE-CI) / methanol to yield secondary amines 7 is described. This allows the solid-phase synthetic transformation of the secondary amine 2 into 7. When th

A novel linker for the solid-phase synthesis of secondary amines based on an intramolecular cyclization was developed. The use of two derivatives of 2-methoxy-4-methylsulfinylbenzyl alcohol is demonstrated as a safetycatch protecting group and linker for solid-phase peptide synthesis. The protecting group and linker are stable to TFA and are readily removed under reductive acidolytic conditions.
