โฆ Synopsis
Sweat (1) reported that many steroids yielded fluorescent products in the presence of concentrated acids. Silber, Busch, and Oslapas (2) and Zenker and Bernstein (3) applied this observation for an analytical procedure for corticosteroids in biological tissues. The specificities of the methods are dependent on the efficient development of fluorophores for a few corticosteroids. There may, however, be difficulties with background or interfering fluorescence, and Flack and Stockham (4) have shown that many biological extracts assayed by the method of Zenker and Bernstein
(3) contain appreciable amounts of cholesterol. The present paper describes a method of correcting for background or interfering fluorescence using cholesterol as an example of an interfering substance in the fluorescence assay of corticosterone. The method is based on that described by Glenn (5) for two-component spectrophotometric analysis.
