β¦ Synopsis
The determination of hydroxyproline has become increasingly important in view of its use in the detection of metabolic bone disease and the measurement of collagen turnover under a variety of experimental conditions. The most commonly used method is the one described by Prockop and Udenfriend (1). In our hands this method failed to give good recoveries and duplicate analyses did not agree. This paper describes the development of a chromatographic procedure for the determination of hydroxyproline in urine and bone extracts. In this method, the mean error of duplicate analyses is only 2oJ0, and recoveries of 98.3% are attainable. In addition, two shorter methods are described that do not involve chromatography. These methods are capable of high precision, but are less accurate. One, the "direct acid procedure," may be suitable for the rapid, routine screening of clinical urine specimens.
