✦ Synopsis
Abstract
The title compounds were synthesized starting from dimethyl (2S,3aR,8aS)‐(+)‐8‐(phenylsulfonyl)hexahydrophyrrolo[2,3‐b]indole‐1,2‐dicarboxylate. This compound on treatment with LDA followed by reaction with [^14^C]methyl iodide or [^3^H]methyl iodide gave the methyl substituted derivative which on treatment with trifloroacetic acid followed by hydrolysis with alkali gave the desired α‐[~14~C]methyl‐ or [^3^H]methy‐L‐tryptophan, respectively, in 50‐60% radiochemical yield.

salts (1)-( 3) are rapidly decomposed by moisture, but (4) can be kept open to the air for several hours since its orthomethyl groups shield the center of the positive charge.
Local cerebral serotonin synthesis capacity was measured with ␣- [C-11]methyl-L-tryptophan ([C-11]AMT) in normal adult human brain (n ϭ 10; five males, five females; age range, 18-38 years, mean 28.3 years) by using positron emission tomography (PET). [C-11]AMT is an analog of tryptophan, the precur
## Abstract Improved procedures for the radiosynthesis of α‐[^11^C]methyltryptophan and α‐[^11^C]methyl‐tryptophan methyl ester were studied. Following α‐deprotonation of tryptophan methyl ester benzaldimine with LDA, 60 – 80% of no carrier added [^11^C]iodomethane was incorporated in 5 minutes at
