In vivo measurements of D-9 desaturation of myristic, palmitic, and stearic acid in lactating dairy cattle A method is described for assessing the activity of D-9 desaturase in vivo. A mixture of the three 1-13 C acids was infused into a cow over 24 hours or infused into the abomasums as a bolus over 20 minutes or 24 hours. Milk fat was recovered, converted to methyl esters, and then to DMDS adducts (dimethyl disulfide) for examination by GC-MS. The desaturase products of the saturated acids formed during continuous infusion of the bolus for 16 hours was found to be 90% of the 14:1, 50% of the 16:1 and 59% of the 18:1. As expected the monoene acids were cis-9 isomers.
