✦ Synopsis
Abstract
Nitrosoguanidine mutagenesis of the Group A streptococcal strain K56 known as a good recipient in phage‐mediated transduction led to the isolation of two recombination deficient mutants showing transduction frequencies of chromosomal markers about 100‐fold lower than those of the parent strain. These mutants were sensitive to ultraviolet (UV) irradiation but had retained their ability to reactivate UV‐irradiated phage A25. They were cross‐sensitive to the killing action of nitrosoguanidine and ethyl methane sulfonate and showed severely inhibited DNA synthesis after UV radiation.

Two mutants deficient in homologous genetic recombination have been isolated from Rhizobium meliloti 41 after Tn5 mutagenesis. Both mutants are defective in the induction of temperate phage 16-3 by UV-light, Mytomycin-C or Bleomycin, their UV sensitivity is more pronounced than that of the wild-type
The isolation of a recombination deficient (Rec-) strain of Agrobacterium tumefaciens is described. Strain LBA 4011 was mutagenized with nitrosoguanidine and after segregation 18,000 colonies were replica plated and UV irradiated. Twentytwo UV sensitive strains were isolated and tested for methylmet
Plasmid pIF132 containing two direct repeats of the mel (melanin) sequence was used to monitor intraplasmid recombination. Five mutants of Streptomyces lividans TK64 deficient in intraplasmid recombination were isolated. Four contained additional defects in aerial mycelium formation, pigmentation, a
