✦ Synopsis


A systematic study of the enhancement of optical absorption of solutions upon freezing is presented. The enhancement factor, the ratio of absorbance of the frozen solution under given conditions to that of the solution at room temperature, is shown to increase with the optical pathlength of the sample, lower temperatures, and decreasing optical density of the solution. The enhancement factor is only weakly dependent upon wavelength under the defined conditions. This study makes possible a clearer understanding of the factors involved in lowtemperature spectroscopy. Also presented are measurements of the relative contributions of the two hemes of cytochrome oxidase to the optical spectrum at -14O"C, as an example of quantitative studies at low temperatures.
