β¦ Synopsis
Ethanol-induced adsorption of amylose on a cellulose column equilibrated with ethanol-urea is used for the quantitative separation of amylose and amylopectin from starch. Under these conditions, amylose is completely retained on the column. The column is washed free of amylopectin and the adsorbed amylose is eluted by gradient elution with ethanol-urea.
Samples containing 300-400 mg of starch can be successfully fractionated on a column packed with defatted cellulose (20 g) with a recovery of 95k2o/o. The elution profile reveals the heterogeneity of native amylose.
