โฆ Synopsis
Background and Objective: Transgenic mouse models of mammary cancer are useful tools for investigating neoplasia. Applying PDD in these animals the heme precursor aminolevulinic acid (ALA) will rapidly metabolize by the tumor cells into the fluorescent protoporphyrin IX (PPIX). Study Design/Materials and Methods: We have tested PDD in multiple mouse models of breast cancer to investigate the generality of this finding and to explore the limits of PDD to demarcate tumor margins. ALA to PPIX conversion was performed following tail vein injection of ALA (100 mg/kg). These tumors arose in transgenic mouse strains with directed expression of oncogenic proteins in the mammary epithelium (HRAS, NEU, Polyoma virus T antigen (PyVT), and SV40 Large T antigen (WAPTag), or via infection with mammary tumor virus. Results: In this strain survey, each tumor could be positively identified macroscopically by moderate to strong PPIX fluorescence intensity under 400 nm light when examined in situ, 1-2 hrs post ALA injection. In addition, we were able to discern a lung metastasis originating from a mammary tumor of a PyVT female, and early hyperplastic lesions in mammary glands of a WAPTag female. Conclusion: Our preliminary experiments have confirmed that strategies for ALA-mediated PDD can be further investigated and optimized in mouse models of breast cancer, and these advances should be directly translatable to human clinical practice.
