β¦ Synopsis
Polymer-shielded dye-affinity chromatography is a form of chromatography in which the dye matrix forms complexes with a nonionic, water-soluble polymer such as poly(viny1pyrrolidone) or poly(viny1 alcohol), prior to the column chromatography of a crude protein extract, the idea being that polymer shielding of the dye will prevent nonspecific interactions between the target protein and the dye. The concept of polymer shielding and a strategy for the rational selection of polymers suitable as shielding agents are presented.

Continuous beds derivatized with three triazine dyes (Cibacron Blue 3G-A, Pricon Red HE3B and Pricon Red H-3B) were used for chromatographic purification of dehydrogenases from yeast enzyme concentrate. A dye affmity fractionation of a commercial pectic enzyme preparation is described. From nine immobilized triazinic dyes assayed, only Blue R-HE and Yellow 4R-HE were able to retain pectin lyase thus allowing the obtention of a fraction not producing methanol during fruit juice clarification. While
