โฆ Synopsis
Measures of adenosine triphosphate (ATP) and other adenosine phosphate compounds are useful indices of microbial biomass and microbiological activity. Various photometric procedures for the determination of these compounds are compared. A modification in the well-known hexokinase-glucose-6-phosphate dehydrogenase procedure for ATP, which permits a very sensitive kinetic measurement of ATP + ADP (-3. I pmol), is presented. The procedure utilizes phosphoenol pyruvate and pyruvate kinase to regenerate ATP (substrate in the preceding reaction) and thus is an amplification system. Longer reaction times and fluorimetric measurement of NADPH produced would further enchance the sensitivity of the procedure. Finally, a simple, rapid far-uv spectrophotometric procedure, previously believed to be specific for adenosine monophosphates, is demonstrated to provide quantitative estimates for adenosine diphosphate (ADP) and ATP as well.
