✦ Synopsis
We read with interest the article by Van Gijssel et al. published recently in HEPATOLOGY, 1 where, employing a Solt-Faber protocol, these investigators show an attenuated nuclear accumulation of p53 in response to DNA damage in preneoplastic lesions in vivo. We would like to point out that there are two previous publications showing a lack of p53 response to genotoxic agents in similar lesions in vivo. 2,3 These studies are not referred to in the report by Van Gijssel et al., but complement their discussion.
In the studies by Lennartsson et al. 2 and Finnberg et al., 3 a different protocol was used to induce preneoplastic hepatocytes, i.e., the genotoxin diethylnitrosamine was administered both as initiating agent and promoter. This protocol also gave rise to preneoplastic lesions with an attenuated nuclear p53 response. 2 The proportion of nonresponsiveness increased with the size of the lesions 2 and with the number of weekly doses, 3 supporting the notion that the hepatocytes localized in
