Abstract
The effect of the Fv locus on the pathogenesis of Friend virusโinduced leukemia was studied with two pairs of mouse strains which are congenic except for the locus. The rapid spleen enlargement or spleen focus formation which is characteristic of Friend virus infection occurred in the mice with Fv^s^/Fv^s^ genotype, DDD and C57BL/ 6โFv^s^. These early responses were not seen in the mice with Fv^r^/Fv^r^ genotype, DDDโFv^r^ and C57BL/6. When the infected Fv^r^/Fv^r^ mice were observed for 150 days, all of the DDDโFv^r^ mice, but none of the C57BL/6, developed splenomegaly. Histological studies of the DDDโFv^r^ mice revealed erythroblastic, lymphatic or granulocytic leukemia, or myeloproliferative disorder with a prominent megakaryocyte proliferation. This observation indicates that Friend leukemia virus is able to induce various types of leukemias in mice and the early characteristic responses are completely abrogated in the mice with Fv^r^/Fv^r^ genotype. Involvement of genetic factor (s) other than the Fv locus in the lateโoccurring leukemias was suggested.
