✦ Synopsis

A number of recent clinical observations suggest that vitamin D 3 plays an important role in maintaining normal cardiovascular function, either directly through its receptor in cardiac muscle, or indirectly through its infuence on circulating levels of calcium or on other regulatory factors. By using an antibody directed against the recombinant vitamin D 3 receptor, we have identified the receptor protein for 1,25(OH) 2 D 3 in tissue from two human hearts. The identification of the 1,25(OH) 2 D 3 receptor in human heart lends credence to the hypothesis that 1,25(OH) 2 D 3 directly effects the human heart and may be involved in several clinically relevant pathological conditions involving the vitamin D 3 endocrine system. 1996 Academic Press Limited Kο₯: vitamin D; receptor; human heart.

In this paper, we detail an enzyme-linked immunoassay for the 1,25-dihydroxyvitamin D 3 receptor protein. The receptor protein of cell and tissue homogenates is bound between two monoclonal antibodies specific for different epitopes on the receptor protein. The first antibody is bound to the well of

In the present investigation we studied the metabolism of 1 cu,25-dihydroxy-[l P-3H]vitamin D3 (3H-1 ,25(OH),D3) in culture-grown human keratinocytes (CHK). O u r results showed that the cellular uptake of 3H-1,25(OH)2D3, upon incubation with CHK, occurred very rapidly; and it paralleled a decrease

Differentiation and proliferation can be regulated in diverse cell types by 1,25-dihydroxyvitamin D 3 . These effects derive from modulation of gene expression mediated by the interaction of 1,25-dihydroxyvitamin D 3 with the vitamin D receptor (VDR). The VDR is one of the nuclear hormone receptors.

1␣,25-dihydroxyvitamin D 3 (1␣,25(OH) 2 D 3 ), the active metabolite of vitamin D, mediates many of its effects through the intranuclear vitamin D receptor (VDR, NR1I1), that belongs to the large superfamily of nuclear receptors. Vitamin D receptor can directly regulate gene expression by binding to
