CryoEM can be divided into three different sub-disciplines: electron crystallography where the macromolecules are arranged periodically into thin crystals, single particle cryoEM of randomly oriented isolated macromolecular assemblies and electron tomography of vitreous specimens containing objects

We describe a temperature-jump device that permits time-resolved studies of thin cryo-transmission electron microscopy specimens. The specimen is rapidly heated to induce a change in microstructure just prior to cryo-fixation. The apparatus consists of a xenon arc lamp equipped with a shutter contro

Dimensional protein crystals imaged at low temperature and under low dose conditions reveal information to better than 3A resolution. Imaging under similar conditions and image reconstruction of non periodic specimens such as spherical viruses have yielded structural detail to only 30A resolution.
