Implementation of human bioavailability studies with the analgesic indoprofen required a rapid, sensitive, and convenient assay in blood plasma. A procedure based on ether extraction of acidified plasma, derivatization with diazomethane, and GLC analysis using a 3% OV-1 column was sufficiently sensitive for measurement of plasma indoprofen concentrations in the 0.4-16 mug/ml range. An average recovery of 99.0 +/- 7.6% (SD) was achieved when the pentanoic acid homolog was employed as an internal standard.

A GLC method for phenylbutazone at concentrations down to 10 ng/ml in human plasma is described. After addition of an internal standard, phenylbutazone is extracted at pH 5 into benzene. The dry extract is dissolved in benzene, and phenylbutazone is determined by GLC using a 63Ni-electron-capture de
