✦ Synopsis

Ehrlich ascites tumor cells spontaneously release cell surface material (glycocalyx) into isotonic saline medium. Exposure of these cells to tritium-labeled 4,4'-diisothiocyano-1,2-diphenylethane-2,2'-disulfonic acid ( H2 DIDS) at 4Β°C' leads to preferential labeling of the cell surface coat. We have combined studies of the kinetics of H2 DIDS-label release, the effects of enLymatic treatment, and cell electrophoretic mobility to characterize the 1-12 DIDSlabcled components of the cell surface. Approximately 7 ted radioactivity is spontaneously released from the cells The kinetics of release is consistent with the first-order loss of t w o fractions; a slow

We have developed a simple and rapid method for isolation of purified nuclear lamina from Ehrlich ascites tumor cells. The procedure employs chromatin structures prepared from whole cells at low ionic strength and is carried out under conditions that minimize the formation of artifactual protein-DNA

The effect of extracellular Pi and arsenate on Pi-transport in Ehrlich ascites tumor cells has been studied. Pi-transport can be described by Michaelis-Menten kinetics; the maximal flux equal to 44 mmoles (kg cell water)-' hour-' and K m equal to 3.3 X 1 0 -4 M. Arsenate is a competitive inhibitor o

The effects of extracellular P, and Na' on cellular Pi concentration and transport were studied. Steady-state P, exchange flux was measured by 3zP uptake in the presence and absence of Na'. Model experiments were also conducted to assess t h e possibility that hydrolysis of organic phosphate esters
