We have used surface plasmon resonance (SPR) to follow variations in the concentrations of binary complexes as RNA polymerase moves into a transcriptionally competent initiation complex with immobilized DNA fibres containing promoter sequences. The use of SPR to follow complex binding phenomena is d

β¦ Synopsis
In vitro transcription in a HeLa cell lysate by RNA polymerase II directed by a chicken feather keratin gene promotor has been studied using unmethylated template DNA and DNA methylated in vitro by HpalI methylase. The efficiency of specific gene transcription from methylated DNA was dependent on topology of the input DNA, the most significant effect being complete inhibition of transcription from one template which contained three methylation sites, one just 5' and two greater than 500 bases 3' to the site of transcription initiation. The inhibition of transcription depends on a factor(s) which is variably present in lysate preparations and is labile on storage at -70 ~

S. typhimurium strain BY324 is temperature sensitive due to a mutation (rpo C32) in the gene for the RNA polymerase fi' subunit. Transcription of T7 DNA by RNA polymerase purified from this strain is temperature sensitive in vitro. The enzyme is slightly defective in template binding and RNA chain i
## Abstract Bacteriophage T3βinduced RNA polymerase is rapidly inactivated at 42Β°C. Addition of T3 DNA delays this process for 30 s and reduces the rate with which the enzyme activity is lsot indicating that a labile binary complex between T3 DNA and polymerase must have been formed. The ternary co
## Abstract We have investigated the role of phosphorylation by vertebrate protein kinase CK2 on the activity of the General Transcription Factors TFIIA, TFIIE, TFIIF, and RNAPII. The largest subunits of TFIIA, TFIIE, and TFIIF were phosphorylated by CK2 holoenzyme. Also, RNA polymerase II was phos
