✦ Synopsis

Abstract

One of the problems in studying early acting recessive lethal genes is recognizing the homozygotes prior to their demise. Molecular probes can assist in this task, but their use generally requires removal of cells and consequent damage to the embryo, which may compromise its subsequent development. The present study was undertaken in an attempt to distinguish intact, living, lethal yellow (A^y^/A^y^) embryos from A^y^/a^e^ and a^e^/a^e^ littermates before implantation by placing them in implantation delay or diapause. After 2 days in the reproductive tract of prepubertal females, the great majority of presumptive lethal A^y^/A^y^ embryos had failed to hatch from the zona pellucida and they exhibited a marked deficiency of cells relative to controls, particularly in the inner cell mass. This argues against a stage‐specific role for the gene in implantation. Β© 1992 Wiley‐Liss, Inc.

Strain C57BL/6J-Ay/Ay embryos are developmentally retarded in utero at 90 hpc and are resorbed by 132 hpc. The Ay/Ay developmental delay previously observed in vitro has now been confirmed in vivo. Apparently Ay is involved in some aspect of cleavage stage metabolism.

## Abstract At present, experimental studies on the primary genetic lesion in A^y^/A^y^ embryos are restricted by the absence of techniques designed to identify A^y^ homozygotes prior to the onset of secondary defects. This paper describes a methodology to enrich the yields of A^y^ homozygotes in e
