A new assay of catechol O-methyltransferase (EC 2.1.1.6) is described by determination of the W-and p-O-methylated products of 3.4.dihydroxybenzoic acid. The method involves DEAE-Sephadex A-25 chromatography and reversed-phase high-performance liquid chromatography on a LiChrosorb 5 RP 18 column.

โฆ Synopsis
In a study of in vitro and in vivo metabolism of L-3,4dihydroxyphenylalanine
(L-Dopa), two methods of high-performance liquid chromatography (HPLC) were used to separate the m-and p-O-methylated products. A reversed-phase column and an aqueous mobile phase by gradient elution were used; the elute was analyzed ekctrochemically with a single amperometric and dual coulometric electrode. The L-Dopa and its 0-methylated products could be detected individually in the enzymatic methylation of rat liver homogenate and in patients with Parkinson's disease. Meta/para ratios of O-methylation are easily obtained by this method. Q 1985 Academic PES, IIIC.

We have developed a liquid chromatographic method which uses electrochemical detection for the simultaneous quantitation of histamine and N'-methylhistamine in rat brain. The amines are derivatized with the water-soluble Bolton-Hunter reagent (sulfo B-H). Perchloric acid extracts of rat brains are c
