Synthesis and Reactions of Biginelli Compounds. Part 10. Synthesis and Aromatization of Dihydropyrimidines Structurally Related to Calcium Channel Modulators of the Nifedipine-Type.

-By employing the "Hideg-modification" of the Biginelli reaction a number of 1,4-dihydropyrimidines (IV) (8 examples) is prepared from the ketoester (I), the isourea (II) (used as hydrogensulfate) and aldehydes (II). They can be readily demethylated to the dihydropyrimidones (V) which can be also obtained by the classical acid-catalyzed Biginelli condensation with urea. Furthermore, oxidative aromatization of (IV) is detailed. -(VANDEN EYNDE,
