A High-Yield Synthesis of Deoxy-2-fluoroinosine and Its Incorporation into Oligonucleotides.

-Diazotization of the deoxyguanosine derivative (I) followed by mild fluorination and selective O-silyl deprotection under the conditions shown provides an improved synthesis of the title nucleoside. It is converted to its 5'-dimethoxytrityl-3'-phosphoramidite derivative which is incorporated into a variety of oligonucleotides with up to 7 nonnatural bases. -(ADIB, A.;
