✦ Synopsis

Dear Sir,

As one of the first results from the Swedish Family-Cancer Database, we described in 1997 a strong familial clustering of breast cancers and melanomas at the population level. 1 In a follow-up study, more such families were identified and the high risks were confirmed. 2 For example, in offspring of mothers with breast cancer, the familial standardized incidence ratio (SIR) for melanoma was 5.70 (CI ϭ 2.59 -10.04) when the father had melanoma, compared to an SIR of 1.14 (CI ϭ 1.02-1.27) when the father had no cancer. The offspring risk for breast cancer in such families was 4.74 (CI ϭ 2.67-7.44

A gene (BRCA1) predisposing for familial breast and ovarian cancer has been mapped to chromosome band 17q12-21. Based on the observation that ovarian tumors from families with breast and ovarian cancer lose the wild-type allele in the region for the BRCA1 locus, it has been suggested that the gene f
