✦ Synopsis


A continuous automatic method involving alkaline hydrolysis, neutralization with sulfuric acid, and specific detection of 4-hydroxyproline is described. The main parameters of the method are studied. This technique is capable of detecting 10 pmoliliter of Chydroxyproline linked in peptidic sequence with acceptable repeatability (SD/mean = 0.05). The automatic alkaline hydrolysis yield is 67% for soluble type I collagen. The method enables the analysis of 500 samples in 24 h.
