โฆ Synopsis
Radioiodine-substituted nucleosides in "3-labeled poly(U) are rapidly deiodinated by reaction with bisuhite at near neutral pH. The deiodination reaction is prevented by annealling of the iodinated poly(U) with poly(A). The latter characteristic is the basis for an analytical protocol used to characterize the composition of poly(A)-RNAs of the rat thyroid. A high molecular weight RNA species sedimenting at a rate similar to that of beef thyroglobulin mRNA was detected. The result of the analysis indicated that the rat RNA corresponding to the beef thyroglobulin messenger can be segregated by sedimentation. The assay for poly(A)-RNA based upon resistance to deiodination is advantageous in the analysis of small amounts of RNA since the test does not result in fragmentation of the polynucleotides.
