โฆ Synopsis
An interesting observation reported in our previous studies (1,2) is that, on binding to nucleic acids (NA) the dye, ethidium bromide (EB) increases its fluorescence quantum efficiency by a factor of 20 to 25. In consequence of the EB absorption spectrum shift on binding, for a suitably chosen wavelength of fluorescence excitation, the intensity of fluorescence emission increases by a factor between 50 to 100 when EB binds to NA.
Furthermore it has been shown ( 1) that under suitable conditions the fluorescence increase of an EB solution of a given concentration is proportional to the concentration of nucleic acid added. These results provide the basis for a new spectrofluorometric method for NA determination, described herein.
