Abstract
A simple method of chemical phosphorylation for modified nucleosides with [^32^P]orthophosphoric acid in the presence of BrCN is described. The yields of 5β²β[^32^P]nucleoside monophosphates achieved are 50β65% at nominal specific radioactivities of ca. 1000βCi/mmol. The mechanism of phosphorylation in the presence of heterocyclic amines is studied. Copyright Β© 2008 John Wiley & Sons, Ltd.

A simple method for preparing s-32P-labeled nucleoside-5'-triphosphates of high specific activity is presented. Unprotected nucleosides are heated dry with urea and inorganic phosphate. The products from such reactions are phosphorylated enzymatically.
Anomeric Ξ±and Ξ²-N,O-psiconucleosides were prepared by 1,3-dipolar cycloaddition of C-ethoxycarbonyl N-methyl nitrone with ethyl 2-acetyloxyacrylate, followed by VorbrΓΌggen nucleosidation. The synthetic scheme has been applied to all purine and pyrimidine nucleobases. Nucleosidation can proceed under
