However, the present method is somewhat simpler with respect to the number of solutions which must be prepared, and, since the velocity is measured by the increase in absorbance at 385 mp, it can be easily modified for use with spectrophotometers not having the ultraviolet attachment. In this case, buffer and substrate can be incubated with enzyme and after a fixed time interval the reaction stopped by the addition of acid. The absorbance at 385 rnp is then read against a blank which is prepared by adding acid to the buffer and substrate prior to the addition of enzyme.
