The present studies were undertaken to obtain a high concentration of renin-substrate.

Hog serum was fractionated by salting-out with graded increments of NaC1 concentrations at pt{ 3.1. A substrate concentration of 30 times that of the one in the original serum--in terms of ng angiotensin produced per minute and per mg protein--was obtained in a fraction resulting from an addition of 180--210 g NaC1 to 1 litre of serum.

A further 30 times purification was achieved through the treatment of the above fraction with Sephadex G-75, whereas the use of Sephadex G-50 or G-25 lead to a lower degree of purification. Thus the combination of the salting-out and the Sephadex G-75 methods resulted in a substrate concentration of approximately 1000 times that found in the untreated serum.

When after salting-out, the Sephadcx treatment was replaced by ultracentrifugation in a graded sugar-density-medium, the degree of purification was lower than with te Sephadex treat ent. However, three concentrat on-maxima at various degrees of sugar density were observed during the ultra-centrifugation, which suggested the presence of at least three different renin-substrates.
