Abstract

A total synthesis of L‐(+)‐swainsonine, a potent and specific inhibitor of naringinase, along with the syntheses of six unnatural indolizidine azasugars are reported by starting from D‐glucose. L‐(+)‐Swainsonine was synthesized in 14 steps in 17 % overall yield. Further, two of the indolizidine analogues were found to be good glycosidase inhibitors at micromolar concentrations. In all of these syntheses, the key step was an intramolecular S~N~2 reaction. (© Wiley‐VCH Verlag GmbH & Co. KGaA, 69451 Weinheim, Germany, 2008)

## Abstract L‐Glucose derivatives are prepared by two different routes. The first involves a modification of the previously reported multi‐step approach by __Shiozaki__^8^, starting from a D‐glucurono‐1,5‐lactone derivative, resulting in the isolation of 2,3,4,6‐tetra‐__O__‐benzyl‐L‐glucopyranose.

Synthesis of cis-3-hydroxy-L-proline from D-glucose is reported. The methodology involves conversion of D-glucose into N-benzyloxycarbonyl-c-alkenyl amine which on 5-endo-trig-aminomercuration gave the pyrrolidine ring skeleton with sugar appendage in 25% yield. Alternatively, N-benzyloxycarbonyl-c-

The total synthesis of (+)-azimic and (+)-carpamic acids are described, based on a biomimetic heterocyclization of optically active precursors derived from D-glucose, The piperidine alkaloids constitute a large family of naturally occurring substances many of which possess important physiological a
