Thin-layer chromatography of keto acid 2,4-dinitrophenylhydrazones was carried out on plate coated with silica gel and sodium bicarbonate. A clear separation was obtained between most of the various keto acid hydrazones. The hydrazone was eluted from the plate with methanol containing 1% concentrated ammonia. A complete recovery was obtained and isomerization of the hydrazones was found not to occur. These results indicate usefulness of the present method for separation, detection, and determination of keto acids. Analysis of keto acids by this method is described for culture fluids of some microorganisms.
